Annotation description: | Expression of VAMP4 was investigated in area CA1 of the hippocampus by immunocytochemistry and immunoelectron microscopy. While prominent immunofluorescence staining was observed in the cell body layer (stratum pyramidale), distinct labeling was also observed in subsets of nerve terminals. Immuno-EM revealed sparse labeling of vesicular structures in presynaptic nerve ending, consistent with its presence on presynaptic endosomes. 5/2/2017 Pim - Based on the known transmembrane topology of the protein, the "integral component of ... membrane" term was chosen. |

Additional literature: | The first subcellular localization study of Vamp4 was done by Steegmaier et. al (1999). Using immunofluorescence and electron microscopy, they showed that VAMP4 localized to tubular and vesicular membranes of the Trans-Golgi network (TGN). Double labeling of VAMP4 and syntaxin 6 showed that these two SNARE proteins colocalize in the same TGN membranes. Also, double-immunogold labeling of VAMP4 and clathrin revealed that VAMP4 is present on clathrin-coated vesicles in the TGN, clathrin-positive Insulin secretory granules and endosomal vacuoles. @ PMID:10359608 |
