Annotation description: | Fig.3: "Cell aggregation assay using NRG-expressing COS7." - "NRGb1-expressing COS7 cells showed a markedaggregation in contrast to the parental COS7 cells, impli-cating a homophilic adhesive activity on the part ofNRGb1. Furthermore, cell aggregation activity ofNRGb1-expressing COS7 cells was inhibited by theNRG antibody, ANB104 (5 mg/ml), that recognizes anIg-like domain of NRG." Fig. 7: "An electron micrograph of the synaptic junction in cerebellar sections. Subcellular localization of NRGb1. Immuno-gold electron microscope analysis using ANB101 (A and B) and ANB102 (C) antibodies revealed that NRGb1 molecules were localized in a glomerulus of the granular layer, either postsynaptically in the dendritic profiles(A), or presynaptically in the mossy fiber terminals (B), and occasionally in the synaptic clefts (C)." Summary: the authors show expression of NRG1 in pontine nuclei (presynaptic neurons) and cerebellar granule cells (postsynaptic neurons). Immuno-EM shows NRG1 localization in pre-and postsynaptic elements located in the granular layer of the cerebellum (has mossy fiber/granule cell synapses). Sometimes gold particles are found in the synaptic cleft. Fig.3 NRG1 is shown to possess homophilic adhesive properties. The model is that some NRG1 is located at the pre- and postsynaptic specialization membrane and has a function in membrane adhesion. |
