Annotation description: | SAP-102 is the dominant PSD-MAGUK responsible for the synaptic targeting of AMPA receptors during early postnatal development. Previous work showed SAP-102 protein is expressed strongly by postnatal day 10 (P10, robust PSD-95 and PSD-93 expression occurs 3 weeks later, Sans et al., 2000). "SAP-102 is not required for synaptic AMPA or NMDA receptor expression at mature synapses." shRNA knockdown of endogenous SAP-102 in pyramidal neurons had no effect on basal synaptic transmission (Fig. 5C1- 5C3). "SAP-102 shRNA caused an ~50% reduction specifically in AMPA-R-mediated synaptic transmission in immature synapses. (Fig. 8G1-8G3)." Furthermore, shRNA knockdown of PSD-95 did not affect synaptic transmission in immature synapses (Fig. 8F1-8F3) supporting the notion that SAP102 is the dominant PSD-MAGUK at this age. Figure 1. shRNAs Against PSD-95, PSD-93, and SAP-102 Cause Selective Knockdown in COS-7 Cells and Hippocampal Neurons. Figure 9. "Slice Culture Scheme" shows how mature vs immature synaptic transmission was assayed in hippocampal slices. |
