Annotation description: | Areas of interest: latero-ventral hippocampus (CA1 radiatum and CA3 lucidum-radiatum), the deep layers (V-VI) of the primary motor cortex Using electron micrographs, EphA4 was detected with anti-EphA4 immunoperoxidase labeling in the adult mouse hippocampus within axon terminals and dendritic spines. Some dendritic shafts were occasionally labeled in the mouse hippocampus. No immunoperoxidase staining was found in EphA4 KO mice. Using pre-embedding immunogold labeling in electron micrographs of CA1, anti-EphA4 silver-intensified immunogold particles (SIGPs) were significantly concentrated in axon terminals (plasma membrane and vesicles). Fig 3. On Western Blots, EphA4 is detected in presynaptic active zone fractions from purified synaptosomes that are positive for SNAP25 and synaptophysin (and negative for PSD95 and GluR2/3) Fig 4. EphA4 is preferentially detected in axon terminals and dendritic spines in area CA3 of hippocampus, using immunoperoxidase staining in EM. Fig 5 and quantification in Fig 6.
