| Annotation description: | From Results section: The 1.0/1.3 M sucrose interface consists primarily of synaptic plasma membranes with a portion of their postsynaptic membrane and postsynaptic density attached. (...) Using the anti-14-3-3 antiserum as a probe, the immunoreactivity was most concentrated in the M1.0 and synaptic plasma membrane fractions. To characterise further the location of the 14-3-3 in the synaptic membrane preparation, the iodonitrotetrazolium-treated membranes were extracted with 0.2% Triton X-100, and the Triton-soluble and -insoluble components were separated by centrifugation through a layer of 1 M sucrose. (...) The ß/ζ isoforms appeared to be quantitatively solubilised by the treatment, whereas the isoforms γ, ε, and η were present in each of the three fractions at similar concentrations . This suggests that some of the γ, ε, and η isoforms may be associated with postsynaptic densities. To investigate further the association of 14-3-3 isoforms with synapses, we isolated synaptosomes using the Percoll gradient procedure. (...) interface 3 contains many small but often slightly damaged synaptosomes as well as plasma membranes ; interface 4 is highly enriched in intact viable synaptosomes, most of which have attached postsynaptic membranes with postsynaptic densities; (...) The ß and ζ isoforms were not detectable in the membranes in fraction 4, whereas the γ and ε isoforms were present. |
