- PTPRD is a known presynaptic protein (see ticket #2248). It is shown to mediate cell-cell adhesion with SLITRK1,2,3 in a cell-adhesion assay (Fig.4A+B). SLITRK1,2,3 expressed in HEKcells is able to induce PTPRD mediated clustering of presynaptic (Synapsin1, VGLUT1, GAD67) but not postsynaptic (PSD-95) markers (Fig.4C+D). PTPRD is mediates only the presynaptic differentiation of Glutamatergic synapses via SLITRK1 and 2 (not SLITRK3).Note that PTPRD KD interferes with presynaptic differentiation by SLITRK1, SLITRK2 and SLITRK3 of GABAergic synapses only.
