Annotated paper:Sharma M, et al. "CSPalpha promotes SNARE-complex assembly by chaperoning SNAP-25 during synaptic activity" Nat Cell Biol. 2011 Jan;13(1):30-9 PMID:21151134
Figure(s):Figure 5, 6
Annotation description:In this paper CSPalpha is shown to regulate the protein stability of SNAP25. This activity is mediated by Hsc70 and SGT.

Fig.5: "CSPα binds to monomeric SNAP-25 through Hsc70."
Fig.6: "The CSPα-Hsc70-SGT complex chaperones SNAP-25 and enhances its incorporation into SNARE complexes in vitro."

- Although the data in Fig.6 show the direct role of SGT-alpha (small glutamine-rich tetratricopeptide repeat protein alpha) in the chaperoning in a cell free system. The relevance for and occurence in vivo is suggested by the IP data in Fig.5. Here SNAP-25 IP resulted in selective CSPα, Hsc70 and SGT or SNARE proteins pulldown from brain homogenates.
- Effects on synaptic transmission is not shown in these or other data since the CSP KO is lethal. A role in SV exocytosis is proposed by the authors. This may be a reasonable inference, but I decided not to annotate to term 'regulation of synaptic vesicle exocytosis'.
