Annotation description: | Background: 1] In heterologous cells, the rat GABRR2 homopentamer shows a sensitivity to CACA and a lower sensitivity to picrotoxin (PMID:15733087, Fig.4/5). 2] In the postnatal brain expression of the ρ2 subunits dominate, with a proportion of 85% of all the ρ subunits (PMID:15617751) This paper: Extrasynaptic GABAc receptors have been shown to mediate spillover inhibition in hippocampal GABAergic synapses (PMID:16420458). This spillover inhibition is sensitive to CACA (see 1]). The authors conclude that this effect is mediated by extrasynaptic homopentameric rho2 GABAc receptors. The homopentameric rho2 GABAc receptors are shown to reside in the postsynaptic neuron in Fig.3 via GABA-application. Abstract: "Activation of GABAC receptors with the specific agonist cis-4-aminocrotonic acid (CACA) suppressed postsynaptic excitability and increased the membrane conductance. The GABAC receptor antagonist 1,2,5,6-tetrahydropyridine-4-ylmethylphosphinic acid (TPMPA), but not the GABAA receptor antagonist bicuculline, inhibited the effects of CACA. GABA-mediated long-lasting depolarizing responses evoked by high-frequency stimulation of local inhibitory interneurons in the CA1 area in the presence of ionotropic glutamate receptor and GABAB receptor blockers were prolonged by TPMPA, indicating that GABAC receptors are activated under these conditions." |
