Annotation description: | Fig.2: "In homogenates prepared from neurons in primary culture, WWL70 inhibited ~50% of the total [3H]-2-AG hydrolysis (Fig. 2d)" In cortical layer 5 pyramidal neurons, inhibition of ABDH6 promotes induction of CB1-dependent long-term depression (eCB-LTD) at excitatory synapses using a subthreshold stimulation protocol (Figure 6). This effect is blocked by the CB1 receptor antagonist AM251 demonstrating the action of 2-AG on the presynaptic CB1 receptor (Figure 6). In cultured neurons, 2-AG hydrolysis under basal or stimulated conditions significantly decreases upon inhibition of ABDH6 activity using WWL70 (Figure 3, Supplementary Figure 4). 15/12/2017 Pim - The retrograde transsynaptic signaling aspect of the endocanabiniod signaling in the preparation featured in this paper has been shown in another paper (PMID:17684555): "Sub-threshold stimulation of pyramidal neurons in layer 2 of this brain area induces eCB-dependent LTD (eCB-LTD) in layer 5 when a non-specific inhibitor of 2-AG hydrolysis, URB602, is added." |
