Annotation description: | Shown is an activity-dependent redistribution of Kv4.2 out of dendritic spines in cultured hippocampal neurons. This redistribution requires NMDAR activation and Ca2+ influx and occurs through clathrin-mediated endocytosis. Glycine-induced long-term potentiation (chem-LTP) resulted in concurrent synaptic GluR1 insertion and Kv4.2 internalization. Whole-cell recordings supported these results, showing that endogenous A-type, but not sustained, K+ currents are reduced after chem-LTP induction. Expression of a Kv4.2 dominant-negative mutant was found to increase mEPSC amplitudes and stimuli that induce Kv4.2 internalization resulted in enhanced synaptic currents. Finally, in hippocampal organotypic slice cultures we show that synaptically induced LTP is reduced by hyperpolarization, suggesting that Kv4.2 internalization contributes to LTP. Activity-dependent regulation of Kv4.2 surface expression thus presents a synapse-specific molecular mechanism for the fine-tuning of dendritic signals.
