Annotation description: | Figure 8; "Myo5a mediates the postendocytic recycling of TrkB-FL and contributes to the BDNF-triggered translocation of TrkB-FL into dendritic spines and sustained downstream signaling. (A) Cultured hippocampal neurons (18-21 DIV) were transfected with GFP before the experiment and treated with monensin (10uM) or TAT-5aExon55-66 (1uM) for 30min, followed by BDNF (50ng/ml) stimulation for 60min. No BDNF was added for the control (CON) group. Then, neurons were fixed and permeabilized. TrkB-FL was stained with anti-TrkB (C-14) antibody and labeled with Alexa Fluor 594, as described in the Materials and Methods. Representative images of two types of TrkB-FL localization are shown: arrows indicate TrkB-FL localization in spines and arrowheads indicate TrkB-FL outside the spine. Magnifications are shown to the right. Scale bar: 5um. (B) Quantitative analysis of the percentage of TrkB-FL puncta in mature spines (mushroom-shaped) as a proportion of all TrkB-FL puncta for the cells shown in A. Graphs show mean±s.e.m. determined from the results of more than three independent experiments (n=90-100 spines from the secondary branches of dendrites of 8-10 neurons for each group per independent experiment). *P<0.05 versus CON group; #P<0.05 (χ2 test)." |
