Annotation description:PORCN controls assembly and stability of hippocampal AMPARs at the level of the ER.

shRNA-knockdown of PORCN in hippocampal neurons significantly reduced AMPAR-mediated currents (>50%, Figure 3A-B) and surface and total AMPAR levels (Figure 5A-C). Function of PORCN is independent of catalytic activity (Figure S3B).

AMPAR complex components are reduced in extrasynaptic and synaptic fractions purified from PORCN KO forebrain tissue (Figure 6B-D).

KO PORCN reduces Endo-H-sensitive and resistant pools of AMPARs in hippocampal neurons (TARPs and CNIH only effect EndoH-resistant, Supplemental Figure 5).

- In figures 7 it is shown that the amount of AMPARs in the PSD is reduced. The term 'regulation of neurotransmitter receptor localization to postsynaptic specialization membrane (GO:0098696)' was not awarded because in figure 6 it is shown that "PORCN knockout reduces synaptic and extrasynaptic AMPAR complexes". The effect on AMPAR levels in the PSD is downstream of the number of AMPARs in the membrane.
