Annotated paper:Takei N, et al. "Brain-derived neurotrophic factor induces mammalian target of rapamycin-dependent local activation of translation machinery and protein synthesis in neuronal dendrites" J Neurosci. 2004 Nov 3;24(44):9760-9 PMID:15525761
Figure(s):fig. 8 and fig. 9
Annotation description:mTOR siRNA transfection reduced mTOR (Fig. 8I) and phospho-p70S6K (Fig. 8J) in dendrites upon BDNF treatment in cortical cultures. Figure 8L shows the intensity of red fluorescence, which reflects phospho-S6K signal, in each dendrite that is measured.

In Figure 9 the authors analysed phosph-4EBP levels in isolated dendrites. Even in isolated dendrites, BDNF increased and rapamycin decreased the levels of phosphorylated 4EBP (Fig. 9A-F), indicating the local activation of mTOR-dependent translation in dendrites.

- The BDNF-induced activation of protein translation is rapamycin-sensitive. Rapamycin is an antagonist of mTOR.
- The phospho state of 4EBP1 is used as a readout of the efffect of mTOR on protein translation.\
- Literal: "BDNF increased protein synthesis in the synaptoneurosomes, a concentrated fraction of presynaptic and postsynaptic components prepared from rat brain" --> shows synaptic context
