Annotation description: | Latrophilin-2 loss in postsynaptic hippocampal neurons cultured from KO mice reduced spine density and staining for excitatory markers but not inhibitory synapses, as shown in Fig. 2B-J. In agreement, Fig. 2K,L shows that postsynaptic loss of latrophilin-2 in cultured hippocampal neurons reduces the frequency and amplitude of excitatory miniature events. Fig. 4 shows that latrophilin-2 is also required in vivo for normal spine numbers and its loss reduces spine numbers in CA1 stratum lacunosum moleculare, where it is expressed, but not in other strata. This region-specific effect on functional synapse assembly is supported by the result that postsynaptic Lphn2 deletion in CA1 impairs excitatory synaptic transmission of entorhinal cortex inputs, shown in Fig. 5. |
