Annotation description: | In figures 3 and 5 the authors use immunogold electron microscopy to localize AP180 in rat synapses. Figure 3 is synapses from CA1 neurone and figure 5 is synapses from cerebellar granule cells on Purkinje neurons. The immunogold labelling localizes to the presynapse near the active zone and their is a pool of AP180 on CCVs. 6/12/2017 Pim - The localization to the presynaptic endocytic zone is inferred from the following information: 1. AP180 has a well established role in clathrin coat assembly during endocytosis in non-neuronal system. 2. Next to the active zone is a (stucturally poorly defined) hotspot of SV endocytosis names the 'presynaptic endocytic zone'. Fig.3B+C hows localization of AP180 to a corresponding region bordering the AZ. 3. Localization among clathrin-coated SV is observed (Fig.3A,D,E) These data suggest that (at least in part of its function lifecycle) AP180 is localized to the presynaptic endocytic zone membrane. |
