Annotation description: | In figures 4, 5, and 7 the authors use immunogold electron microscopy to localize CALM in rat synapses. Figure 4 is synapses from CA1 neurone and figure 5 is synapses from cerebellar granule cells on Purkinje neurons. Figure 7 also uses the granule cell synapses. The immunogold labelling localizes to the pre synapse and figure 7 shows a pool on clathrin-coated vesicles. 6/12/2017 Pim - The localization to the presynaptic endocytic zone is inferred from the following information: 1. PICALM has a well established role in clathrin coat assembly during endocytosis in non-neuronal system. 2. Next to the active zone is a (stucturally poorly defined) hotspot of SV endocytosis names the 'presynaptic endocytic zone'. Fig.4K hows localization of PICALM to a corresponding region bordering the AZ. 3. Localization among clathrin-coated SV is observed (Fig.4C,D,...) These data suggest that (at least in part of its function lifecycle) PICALM is localized to the presynaptic endocytic zone membrane. |
