Annotation description:Fig 5 shows that upon Septin 11 knockdown the number of dendritic protrusions is decreased in hippocampal neurons while their length is increased (Fig5B). This suggests a defect in mature spine formation. That this morphological difference also results in less synapses is shown for GABA, Fig5D shows that the number of postsynaptic GABA receptor clusters is decreased upon Septin 11 knock down, and also the overlap between GAD (presysnapse) and gamma2 (postsynapse), indicating that less synapses are formed. This is fully restored by reexpressing Septin 11, and not found in the shRNA with mutations that should not bind to Septin 11 message. These results are further verified by overexpression of a dominant negative Septin 11 that is linked to mCherry. The untagged Septin 11 overexpression has no effect (fig6A-E).
