Annotation description: | IP3R1 was found in postsynaptic ER by immunolabeling of rat cerebellar slices previously embedded in an agarose matrix. 14/3/2017 Pim - Authors use an antibody to "the IP3R" receptor produced in (Mignery et al., 1989, PMID:2554146). There are multiple IP3R genes known today (in mouse 3: Itpr1-3). The antibody was generated to aa 482-500 (llghpphmnvnpqqpa-some aa's missing) of clone pcd 6035 (traced back to accession AAA88319.1). This sequence was blasted against the mouse refseq database. This region does show no detectable sequence similarity with the with the mouse and rat Itpr2 and Itpr3 genes. Results BLAST search: mus Itpr1: 3-11 mus Itpr2: NA mus Itpr3: NA rat Itpr2: 3-15 evalue rat Itpr2: 2.3 evalue rat Irpr3: NA - I approve annotation against rat Itpr1 using the featured antibody generated on mouse sequence. |

Additional literature: | Ultrastructural localization of IP3R1 receptor in mouse cerebellar Purkinje cells by immunogold using three different antibodies against IP3R1. IP3R1 is localizaed mainly in smooth endoplasmic reticulum (ER) (especially, on the stacks of flattened smooth ER, subsurface cisterns and spine apparatus), scantily on the rough ER and on the outer nuclear membrane. @ PMID:2168812 Original study where IP3R1 was found in postsynaptic ER in Purkinje neurons by electron-microscope immunocytochemical localization in rat cerebellar cortex using pre-embedding avidin-biotin labelling @ PMID:2542801 |
