Annotation description: | Authors address the role of Chmp4 indirectly while investigating the role of Chmp2b. They showed that depletion of Chmp2b by means of a shRNA induced a reduction of the density of postsynaptic spines, an effect that was mimicked by a Chmp2b construct unable to bind endogenous Chmp4b (Chmp2b R19/22/26A) which suggests the involvement of both proteins in the process. In particular, this construct was unable to rescue the reduction in the number of spines induced by the loss of Chmp2b in primary hippocampal cultures, while WT Chmp2b was able to fully rescue the phenotype. 18/11/2017 Pim - The annotation of Chmp4b is indirect, but convincing: 1] Chmp2b and Chmp4b are known elements of the endosomal sorting complex required for transport-III (ESCRT-III) complex. Also shown in Fig.8. 2] Fig.3: Chmp2b colocalizes with Chmp4b in dendites in developing neurons. KD of Chmp2b perturbes dendritic morphology. This effect can be rescued by EO of WT Chmp2b, but not by a mutant Chmp2b unable to bind Chmp4b. This shows functional relevance in dendrites. 3] Fig.4: in mature neurons KD of Chmp2b perturbes spine morphology. This effect can be rescued by EO of WT Chmp2b, but not by a mutant Chmp2b unable to bind Chmp4b. - The Chmp4b protein is not easily found in Uniprot. The human Chmp4b protein sequence was blasted against Uniprot and the best matching sequence (97.3% identical) was "Chromatin-modifying protein 4B-like 1 (Rattus norvegicus)" with accession D4A9Z8.
