Annotation description:Authors generated a transgenic mice expressing the auto inhibitory domain in the C terminus of CNAalpha with the rtTA system. These animals were crossed with mice expressing rtTA in neurons under the control of the CaMKIIalpha promoter. Treatment with doxycycline induced the expression of the inhibitory domain of CN and decreased the CN activity in hippocampus and cortex.
Induction of LTP in the Schaffer collateral pathway from hippocampal slices prepared from those mice revealed increased LTP compared to control, in agreement with a role of CN works as an antagonist of LTP. This effect was dependent on PKA activity. Because PPF, which is an index of release probability, was facilitated in mutant animals, the effect on LTP is believed to be caused by postsynaptic mechanisms.
The enhancement of LTP was also reproduced in vivo using anesthetized and awake mice. In this case, LTP was assessed in the Schaffer collateral pathway but also in dentate gyrus.
