Annotation description:Paper characterized PCDH-17 in the corticobasal ganglia circuits with a particular focus on the excitatory prefrontal cortex striatal projections and the inhibitory striatal globus pallidus projections. Utilizing an antibody, whose molecular determinants are not well described, they show excellent specificity utilizing loss of immunoreactivity in western blots, histology and electron microscopy performed with knockout tissue. These antibodies are used in super-resolution immunofluorescence and quantitative immunoglold electron microscopy to show PCDH17 is concentrated in synaptic specialization at the periphery of the active zone and the postsynaptic density on both sides of the synapse. This is true at both the excitatory and inhibitory synapses. The knock-out animals allow investigation of synaptic function and the best evidence is that genetic deletion leads to a reorganization of pre-synaptic vesicles, which in the paper focuses on excitatory synapses. These morphological data hint at an increase in vesicle numbers and this is consistent with the modest reduction in the rate synaptic depression in the PCDH-17 knock out animals relative to the control.
