Additional literature: | Fig. 5A-C, G and Fig. 6: By means of immunoprecipitation of endogeneous neuronal tomosyn and immunostainings in hippocampal neurons, Cazares et al demonstrate the formation of a tomosyn-1, Rab3a-GTP and synapsin 1a/b complex, which is suggested to orchestrate SV partitioning in different SV pools. However, additional experimental evidence for the association of these vesicular peripheral proteins with synaptic vesicles is required since by IPs, no synaptophysin co-enrichment is detected, although colocalization by immunofluorescent co-labelling is observed (Fig. 1 A1-A2). @ PMID:27807164 Geerts et al study the subcellular localization of tomosyn 1 by means of immunofluorescent and immunogold EM staining in neurons. The authors show colocalization with synaptic vesicle markers such as synaptotagmin-1 and VAMP-2 and quantify the fluorescent signal overlap (Fig. 2). Furthermore, they demonstrate association of the protein to SVs and DCV by immunogold labeling (Fig. 2 N-Q). @ PMID:28746398 |
