Rab5 labels endosomal vesicles in Drosophila presynaptic terminals. In Rab5 mutants, these structures are largely missing in CNS and NMJ presynapses. In addition, dominant-negative Rab5 mutant (S43N) localise diffusely in the presynaptic cytosol, concomitant with the accumulation of 70 nm vesicles. These vesicles are thought to be endocytic intermediates that were unable to fuse with early endosomes. The observation is is partially supported by FM1-43 labelling experiments where a significant decrease in labelling of the recycling SV pool was observed in Rab5(S43N) expressing neurons after the initial loading step, suggesting a defect in the endocytic recycling step. @ PMID:12743108
