Annotation description:Authors detect MAPK immunoreactive molecules in PSD fractions prepared from cerebral cortex, hippocampus and cerebellum. Two bands are immunoreactive, which are considered to be ERK1 (43 KDa) and ERK2 (41 KDa). When looking at different subcellular fractions, authors mentioned that ratios of ERK1/ERK2 in the synaptic fractions are different and only ERK2-type MAPK was detected in the PSD fraction.
Authors used a purified Xenopus MAPK to search for ERK2 substrates in different subcellular fractions and compared it with a see start MAPK (p44). All fractions contained substrates for both types of MAPK but Xenopus MAPK (ERK2 type) acted more in substrates in the PSD fraction while the ERK1 type acted more in the soluble and P1 fractions, supporting a role of ERK2-type MAPK in the postsynaptic site.
