Annotation description:GEP100/BRAG2/IQSEC1 is a guanine nucleotide exchange factor for Arf6 at excitatory postsynapses. BRAG2 interacts with AMPA receptors (Fig. 1) and can be co-immunoprecipitated with GluA2 from neurons (Fig. 1E). Subcellular fractionation demonstrates an enrichment of BRAG2 in the PSD fractions, similarly to PSD-95 (Fig. 2B, C). BRAG2 mediated activation of Arf6 is necessary for the internalization of AMPA receptors during chemical (mGluR-mediated; Fig.6, 7) and electrical (NMDAR-mediated; Fig.8) LTD at Schaffer Collateral CA3-CA1 synapses in acute hippocampal brain slices. Knock-down using BRAG2-RNAi and Knock-out (KO) using Cre-infected conditional BRAG2 KO mice prevents the expression of LTD.
In summary, BRAG2 is essential for the expression of hippocampal LTD, which is mediated by the endocytosis of AMPA receptors.

The specificity of the BRAG2 antibody was demonstrated by Western Blot using KD and KO controls (Fig. S3 and S4).
