Annotation description: | IQ-ArfGEF/BRAG1/IQSEC2 is present in the synaptic PSD fraction (Fig.6E) and interacts with PSD-95 (Fig. 6B, C, F) in in vitro-experiments and in homogenates from adult mouse forebrains. IQSEC2 immunoreactivity co-localizes with the labelling of PSD-95, a component of the postsynaptic density of asymmetric, excitatory synapses, but not with that of GABAARbeta1, indicating a specific localization of IQSEC2 to excitatory synapses (Fig.7, 8). Postembedding-immunogold labelling for electron microscopy confirms the presence of IQSEC2 at postsynaptic densities at asymmetrical synapses (Fig. 9). Rabbit antibodies were generated in this study using GST and MBP fusion proteins of the C-terminal region (116 aas) of IQSEC2. The specificity of the antibody was tested by Western Blot (Fig 6D) and using antibody, pre-absorved with the antigen, for IHC (data not shown). |
