Annotation description: | SynArfGEF/BRAG3/IQSEC3 localizes to postsynaptic specializations of symmetrical, inhibitory synapses. Immunohistochemical analysis of the mouse forebrain, cerebellum, and spinal cord revealed that IQSEC3 colocalizes with gephrin, a component of inhibitory postsynaptic specializations (Fig. 1e-g, o-q; s-u; Fig.4b-d, k-n), but not with PSD-95, a component of the postsynaptic density of asymmetric, excitatory synapses (Fig.1h-i). Further, IQSEC3 immunoreactivity colocalizes with the staining pattern of GABAARs (Fig.4e-g) and GlyRs (Fig.4k-n), but not of GluA2Rs (Fig.h-j). Pre-embedding immuno-EM confirmed the presence of IQSEC3-immunoreactivity at postsynaptic specializations of symmetrical synapses (Fig.5). Antibodies (rabbit and guinea pig) were generated in this study using a GST-IQSEC3 fusion protein and specificity was tested by Western Blot (Fig.2) and for IHC by pre-absorption of the antigen (Fig.3b). |
