Annotation description: | Latrophilin-3 regulates the strength of synaptic input from L2/3 to L5 as shown by knockdown of latrophilin-3 in L2/3 neurons and electrophysiological measurements in acute slices (Fig. 1). Specifically, latrophilin-3 regulates strength of synaptic input, not by changing release probability (Fig. 2) but by controlling the density of synapses formed by L2/3 axons in L5 (synapse density in L5 was reduced when latrophilin3 was lost in L2/3 neurons; Fig. 3). The latrophilin-3 Olfactomedin domain was required for binding to FLRT3 (Fig. 5), and mutant latrophilin3 lacking the Olfactomedin domain was not capable of rescuing the deficit in presynaptic density following knockdown of endogenous latrophilin-3 (Fig. 6) |

Additional literature: | Latrophilin-3 regulates glutamatergic synapse number: excess soluble ecto-LPHN3-Fc protein applied to dissociated hippocampal cultures to competitively disrupt endogenous LPHN3 complexes resulted in decreased number of glutamatergic synapses on dentate granule cells by immunofluorescence (Figure 3A-C), corroborated with culture-wide knockdown of LPHN3 that reduced the frequency of mEPSCs (Figure 3K) without affecting mEPSC amplitude (Figure 3L). @ PMID:22405201 |
