Annotation description: | Activation of the G-protein coupled adenosine A1 receptor by adenosine causes an inhibition of neurotransmitter release by modulating presynaptic calcium currents. Principal cells in the MNTB in acute brainstem slices were analyzed by whole cell patch clamp electrophysiology and presynaptic action potentials were evoked by fiber stimulation. Bath application of adenosine caused an attenuation of EPSCs and the effect could be rescued by the application of the adenosine A1 receptor antagonist CPT (Fig.1) indicating that activation of A1 receptors results in an inhibition of neurotransmitter release. Whole-cell voltage-clamp recordings from presynaptic calyx of Held terminals in response to a strong depolarization pulse revealed that bath application of adenosine resulted in an inhibition of presynaptic calcium currents (Fig.4), indicating a clear presynaptic effect for A1 receptors in the calyx of Held synapse. |
