Annotation description: | Authors use electrophoretic mobility shift assays (EMSAs) to detect the presence of kB-DNA-binding activities in the rat brain and rat synaptosomal preparations. They used a 32P-oligonucleoutide containing an NF-kB binding motif which was incubated with cell extracts and subjected to electrophoresis after the binding reaction to detect changes in motility. kB-binding activity was found in synaptosomal fractions with, but also without though less, treatment with DOC, which reveals latent kB-binding activities sequestered by IkB proteins. Previously, authors have described that p65/RelA complexes are responsible for kB-binding activities found in rat brain since incubation the IkB protein MAD3, which inhibits DNA-binding complexes containing p65 abolished the binding activities. The same effect was found when they used an antibody against the N terminus of p65, which also prevented DOC-activated complexes. |
