Annotation description: | The paper makes use of a conditionally-floxed beta-catenin CKO mouse line. The conditional deletion is achieved via a CaMKII-cre transgenic allele (this results in a deletion at around P18). Figure 2: electron microscopy of stratum radiatum synapses shows: - a 25% increase in the number of excitatory, spine synapses (2A,B,D) in the CKO. - a 3-fold increase in perforated synapses (2C,E) in the CKO. - a 40% reduction in the undocked pool of vesicles per synapse (2F) in the CKO. - The number of docked vesicles located less than one vesicle diameter from the presynaptic membrane was similar (Figures 2A, 2B, and 2F). - The reduction in the number of vesicles per synapse in the CKO is not due to redistribution of vesicles over a larger number of synapses. Quantification of the total number of vesicles (in a defined region) shows a 30% reduction in the total number of vesicles. Taken together these data suggest a presynaptic role of beta-catenin in vesicle clustering at the reserve pool region adjacent to the active zone. |
